Severe airway mucous cell metaplasia with highly elevated MUC5AC by immunochemistry and real-time PCR; MUC5B elevated but not statistically significant. |
Evidence
exposure to wood smoke for 3 months induced severe airway mucous cell metaplasia, whereas the control animals did not exhibit mucous cell metaplasia
States the in vivo contrast this assay records: wood smoke produced severe airway mucous cell metaplasia while control animals did not.
Furthermore, both immunochemistry (Fig. 1C,D and H) and real-time PCR (Fig. 1J) revealed that MUC5AC levels were highly elevated in the wood smoke group.
Names the two readouts (immunochemistry and real-time PCR) behind the elevated MUC5AC recorded for the wood smoke output record.
MUC5B expression (Fig. 1E,F,I and J) was also elevated after wood smoke challenge, although this changes was not statistically significant.
Supports recording the MUC5B rise as non-significant rather than as an effect.

Evidence
Interestingly, AR neutralization suppressed most of the WSPM2.5-induced EGFR and ERK phosphorylation (Fig. 7A,B and C) and MUC5AC gene expression (Fig. 7D), whereas blockade of TGF-α or HB-EGF showed a partial reduction, but this decrease failed to reach statistical significance.
States the contrast between AR neutralization and TGF-alpha/HB-EGF blockade that the three treated output records here capture.
Furthermore, recombinant AR dose-dependently induced MUC5AC expression (Fig. 7E). Thus, AR is crucial in mediating the WSPM2.5-induced activation of the EGFR-ERK pathway and MUC5AC up-regulation.
Supports the recombinant-AR arm and the conclusion that AR drives EGFR-ERK activation and MUC5AC up-regulation.
Amphiregulin (AR) was identified as the major ligand responsible for EGFR-ERK signaling activation and MUC5AC expression.
The paper's own summary of the finding this assay records: AR is the major ligand responsible for EGFR-ERK activation and MUC5AC expression.

EGFR-ERK dependence and de novo synthesis of amphiregulin in NCI-H292 cells
Pretreatment with EGFR-neutralizing antibody (0.5 ug/ml), AG1478 (10 uM) or PD98059 (5 uM) significantly reduced WSPM2.5-induced AR mRNA expression and AR release, and exogenous AR (1,800 pg/ml) increased AR mRNA in an AG1478- and PD98059-sensitive way. Actinomycin D and cycloheximide (10 ug/ml) inhibited AR secretion and attenuated MUC5AC mRNA, showing de novo AR synthesis is required. This closes the positive feedback loop between AR release and EGFR-ERK activation. Results are reported as figure panels, so no numeric values are recorded.

Actinomycin D and cycloheximide inhibited AR secretion at baseline and upon WSPM2.5 stimulation, and attenuated MUC5AC mRNA expression. |
Evidence
Pretreatment with anti-EGFR, AG1478 or PD98059 significantly reduced WSPM2.5-induced AR mRNA expression (Fig. 9A) and AR release (Fig. 9B), indicating that the autocrine effects of AR are mediated by the EGFR-ERK pathway in epithelial cells.
States the inhibitor result recorded in the EGFR-ERK-blocked output records: AR expression and release depend on the EGFR-ERK pathway.
ActD and CHX not only significantly inhibited AR secretion at baseline or upon WSPM2.5 stimulation (Fig. 8A and B) but also attenuated MUC5AC mRNA expression (Fig. 8C and D), indicating that de novo synthesis of AR is needed for the transcriptional regulation of MUC5AC.
Supports the ActD/CHX output record and the conclusion that de novo AR synthesis is needed for MUC5AC transcription.
In turn, EGFR-ERK pathway activation was found to contribute to the de novo synthesis of AR.
The paper's own summary of the feedback arm this assay records: EGFR-ERK activation contributes to de novo AR synthesis.

Key-event relationships
EGFR activation leads to mucin hypersecretion
Furthermore, pretreatment with the selective ERK kinase inhibitor PD98059 (Fig. 5C) significantly inhibited WSPM2.5-induced MUC5AC expression, indicating that WSPM2.5-induced MUC5AC production is dependent on ERK activation.
Inhibitor reversal: blocking ERK downstream of EGFR abolished the MUC5AC induction, which is the causal link this relationship asserts.
In summary, this study found that WSPM2.5 induces MUC5AC expression through the AR-EGFR-ERK pathway and that subsequent activation of EGFR-ERK leads to autocrine effects of AR, with the released AR further promoting its own expression.
The paper's own summary that WSPM2.5 induces MUC5AC expression through the AR-EGFR-ERK pathway.

TRIzol RNA extraction; 500 ng RNA reverse transcribed in 10 ul with the PrimeScript RT Reagent Kit (Takara). 2 ul cDNA amplified in 25 ul with SYBR Premix Ex Taq on an MXP3000 QPCR system (Stratagene): 95 C 30 s; 40 cycles of 95 C 5 s and 60 C 30 s; 95 C 1 min; melt 60-95 C. Relative amounts calculated by the 2^-ddCt method.
MUC5AC ELISA on culture supernatants
50 ul of sample incubated with 50 ul bicarbonate-carbonate buffer for 2 h at 37 C, blocked with 2% BSA for 1 h, incubated with mouse anti-MUC5AC (1:200) for 2 h at 37 C, then HRP-conjugated anti-mouse IgG (1:5000) for 1 h. Developed with TMB, stopped with 2 M H2SO4, absorbance read at 450 nm. Standard curve from serially diluted bovine submaxillary gland mucin. Data expressed as the ratio of the observed value to the control group value.
ELISA for EGFR ligands AR, TGF-alpha and HB-EGF
Commercial ELISA kits for AR, TGF-alpha and HB-EGF (R&D Systems) used on cell culture medium per the manufacturer's instructions; optical density measured at 450 nm.
Western blot for phospho-EGFR and phospho-ERK
RIPA lysis with protease and phosphatase inhibitor cocktails, 30 min on ice; centrifuged 12,000 x g, 4 C, 20 min. Protein quantified by BCA. 30 ug protein per lane on 10% SDS-PAGE, transferred to PVDF, blocked in 5% milk, probed 16 h at 4 C with anti-phospho-ERK1/2 (1:1000) or anti-phospho-EGFR (1:10000), then HRP-conjugated secondary IgG and SuperSignal West Femto substrate. Membranes stripped and re-probed with anti-ERK (1:2000), anti-EGFR (1:100,000) and anti-GAPDH (1:1000) for normalization.
MUC5AC immunofluorescence on differentiated ALI cultures
Differentiated bronchial/tracheal epithelial cells fixed in 4% paraformaldehyde 20 min, permeabilized with 0.5% Triton X-100, blocked with 3% BSA 1 h, incubated overnight at 4 C with mouse monoclonal anti-MUC5AC (1:100), then Cy3-conjugated anti-mouse IgG (1:200) for 1 h; nuclei stained with DAPI 5 min. Inserts excised, mounted and imaged by confocal microscopy; MUC5AC signal expressed as the ratio of Cy3 to total DAPI fluorescence (Image-Pro Plus 6.0).
